Kary Banks Mullis (28 December 1944, Lenoir, North Carolina, to 7 August 2019, Newport Beach, California) was an American biochemist who conceived the polymerase chain reaction in 1983 while working at Cetus Corporation, and first demonstrated it successfully that December. The technique let researchers amplify a chosen stretch of DNA into millions of copies from a vanishingly small starting sample, and it remade molecular biology, genetic testing and forensic science; Mullis shared the 1993 Nobel Prize in Chemistry with Michael Smith for the invention, and received the Japan Prize the same year. In the years that followed, Mullis took public positions well outside his own field of expertise and outside the evidence: he questioned whether HIV causes AIDS despite conducting no research on the question, a position that fed into the AIDS denialist policies pursued by South African President Thabo Mbeki's government and that researchers have linked to an estimated 330,000 preventable deaths and 35,000 HIV positive births in South Africa between 1999 and 2008; he also expressed skepticism of human caused climate change and professed belief in astrology and paranormal encounters. Scientists have described this pattern, an accomplished specialist making unsupported claims far outside the field that earned their recognition, as an instance of what is sometimes called Nobel disease.

Kary Mullis has told the story of conceiving the polymerase chain reaction while driving a mountain road in Mendocino County, California, one night in 1983, working through the chemistry in his head as he drove: a way to make a short piece of DNA copy itself over and over, doubling with each cycle, until a single molecule became billions. Whatever the precise circumstances of that night, the idea belonged to real, ongoing work: Mullis was employed at Cetus Corporation in Emeryville, California, a biotechnology company doing exactly the kind of DNA synthesis and enzymology research the technique would need, and the record shows he first demonstrated PCR working successfully on 16 December 1983.

The technique itself is almost absurdly simple to describe and was, at the time, genuinely difficult to make reliable. Heat a DNA sample enough and its two strands separate; cool it and short synthetic primers can find and bind their matching sequence; warm it partway and an enzyme extends those primers into full copies of the target region. Repeat the cycle and the copies double each time, so twenty or thirty cycles turn a sample too small to detect into one large enough to sequence, test or match. Cetus's early instruments cycled the temperature by hand, moving tubes between water baths, until Taq polymerase, an enzyme isolated from bacteria that live in hot springs, replaced ordinary enzymes that fell apart at the temperatures the reaction needed and let the whole process run automated in a single machine.

Mullis shared the 1993 Nobel Prize in Chemistry for the invention, alongside Michael Smith, who was honored separately for a different DNA technique. PCR is now so embedded in molecular biology, genetic testing, forensic science and medical diagnostics that it is easy to forget it began as one researcher's idea about how a molecule that already knew how to copy itself might be made to do so on command, in a laboratory, on a schedule a scientist could control.

This atlas's own standard for a claim is evidence first, and Kary Mullis's later career is a useful, uncomfortable test of what that standard means in practice, because the same man produced both a landmark of twentieth century evidence based science and, afterward, a set of public claims that evidence plainly did not support.

Mullis did no research into HIV or AIDS. He nonetheless argued publicly that HIV had not been shown to cause AIDS, a position virology's own accumulated evidence, built over decades by researchers working in the field, did not support, and the mainstream scientific and medical consensus rejected. That argument did not stay confined to debate: South African President Thabo Mbeki cited AIDS denialist reasoning of exactly this kind in shaping his government's HIV and AIDS policy in the early 2000s, delaying antiretroviral treatment access, and researchers have since estimated the policy contributed to roughly 330,000 preventable deaths and 35,000 HIV positive births in South Africa between 1999 and 2008. Mullis also expressed skepticism of human caused climate change and described paranormal experiences, including an encounter he said involved a glowing raccoon, publicly and without irony.

None of this touches the polymerase chain reaction itself. PCR's validity does not rest on Mullis's later credibility; it rests on the fact that the technique has been independently replicated, refined and verified by tens of thousands of researchers across four decades, in a way his HIV and climate claims never were and never sought to be. Scientists have a name for the pattern his career illustrates, an accomplished specialist trading on real authority in one field to make unsupported claims in others: Nobel disease. The name is only half serious. The consequences, in Mullis's case, were not.
